Located in the cytoplasm, the INK4 family inhibitors sequester CDK4 and CDK6, and prevent the association of the kinase with its cyclin partner, and by extension the phosphorylation and inhibition of RB by the CDK complexes. SERTAD1 can counteract the inhibition of p16INK4a by binding to the complex (Ortega et al. 2002).
